Effects of oxytocin (OT) treatment on the growth of (A) hTERTmyo and (B) SK-UT-1 cells. Cell growth was evaluated by means of an MTS-based assay of cells maintained in 10% serum and treated with OT at a final concentration of 10−6 mol/L for 24 ,48, 72, and 96 hours. All of the experiments were performed in sestuplicate. The data were analyzed by means of a Student t test. ∗P≤.05; ∗∗P≤.01; ∗∗∗P≤.001. FACS analysis of (C) hTERTmyo and (D) SK-UT-1 cells labeled with annexin V and propidium iodide; the cells were untreated, treated for 12 hours with staurosporine at a final concentration of 2.5 × 10−7 mol/L, or treated for 72 hours with OT at a final concentration of 10−6 mol/L.

Expression of oxytocin receptor (OTR) and desmin mRNA in paired leiomyoma and myometrial cells evaluated by means of real-time quantitative polymerase chain reaction. Matched myometrial and leiomyoma cells from two patients were tested at culture passages (P) 1, 3, 5, 7, and 10. Total RNA was extracted and the number of OTRs quantified using HPRT-1 as an endogenous control. The data were analyzed using the comparative Ct method and are expressed in terms of relative expression, which corresponds to 2−▵CT. P3, P5, P7, and P10 values were calculated as percentage of 2−▵CT at P1.

Expression of oxytocin receptor (OTR) mRNA in leiomyoma and myometrial paired samples obtained from 11 patients (numbered from 1 to 11). Patients were in the secretory phase of the menstrual cycle with the exception of patients #4 and #10 who were in the proliferative phase. The total RNA isolated from the cells at passage 1 was quantified by means of real-time polymerase chain reaction analysis using HPRT-1 as an endogenous control. The data were analyzed using the comparative Ct method and are expressed in terms of relative expression, which corresponds to 2−▵CT.
