(A) PHLPP1 regulates Akt2 and Akt3 to terminate signaling to FoxO1, GSK-3α/β, TSC2, and HDM2. PHLPP2 regulates Akt1 and Akt3 to terminate signaling to FoxO1, GSK-3β, TSC2, and p27Kip1. Activation loop phosphorylation of Thr308 in Akt1 corresponds to Thr309 in Akt2 and Thr305 in Akt3. Hydrophobic motif phosphorylation of Ser473 in Akt1 corresponds to Ser474 in Akt2 and Ser472 in Akt3. (B) The phosphorylation status of Akt's hydrophobic motif regulates Akt activity toward the FoxOs and BAD but is dispensable for activity toward GSK-3α/β and TSC2.
