(A) Model in which HELB is recruited to RPA-coated ssDNA behind the resection machinery. In this situation, 5′ to 3′ translocation would lead HELB away from the nuclease ensemble. “B” indicates BLM, “H” indicates HELB, and the yellow pacman indicates EXO1 or DNA2.
(B) Model in which HELB is recruited to the 5′ flap after unwinding but before strand cleavage, thus positioning it to translocate toward BLM and its associated nucleases.
