2 Figure 1 The effect of CMA or DEX on COX-2 mRNA abundance in endometrial explants. Unstimulated and IL-1β–stimulated (1 ng/mL) endometrial explants were exposed to CMA and DEX (10−6 M, each) for 6 hours (n = 15), and the effects on COX-2 mRNA abundance are shown in a box whisker chart. Controls were treated with 0.1% ethanol (unstimulated) or 1 ng/mL IL-1β in 0.1% ethanol (IL-1β). For comparison, the mean COX-2 mRNA levels in uncultured samples are shown. Results are presented as normalized ratios related to the three ubiquitously expressed reference genes G6PDH, ALAS1, and PGK1. Boxes represent lower and upper quartiles with median, whiskers 5th and 95th percentiles. *Significant compared with “uncultured” (P<.05); **significant compared with “unstimulated” (P<.05); ***significant compared with “IL-1β” (P<.05). 3 Figure 2 The effect of CMA and DEX on PGF2α release in endometrial explants. Unstimulated and IL-1β–stimulated (1 ng/mL) endometrial explants were exposed to CMA and DEX (10−6 M, each) for 6 hours (n = 15) in culture before the supernatant was removed and analyzed by enzyme immunoassay (EIA). The effects of the two compounds on PGF2α are shown in a box-whisker plot. Controls were treated with 0.1% ethanol (unstimulated) or 1 ng/mL IL-1β in 0.1% ethanol (IL-1β). The PGF2α concentrations are presented in pg/mg wet weight. Boxes represent lower and upper quartiles with median, whiskers 5th and 95th percentiles. *Significant compared with “unstimulated” (P<.05); ***significant compared with “IL-1β” (P<.05). 4 Figure 3 The effect of CMA or DEX on ANXA1 mRNA abundance in endometrial explants. Unstimulated and IL-1β–stimulated (1 ng/mL) endometrial explants (n = 15) were exposed to CMA and DEX (10−6 M each) for 6 hours in culture. Controls were treated with 0.1% ethanol (unstimulated) or 1 ng/ml IL-1β in 0.1% ethanol (IL-1β). The two compounds were not able to elicit an effect in the tissue samples, and the results have been plotted as a box-whisker plot. Relative ANXA1 mRNA concentrations were significantly elevated in all cultured samples compared with uncultured controls (P<.05). Results are presented as normalized ratios related to the three ubiquitously expressed reference genes G6PDH, ALAS1, and PGK1. Boxes represent lower and upper quartiles with median, whiskers 5th and 95th percentiles. *Significant compared with “uncultured” (P<.05).
