2 Figure 1 Localization of CDC25 in different stages of the corpus luteum. Immunohistochemical staining with anti-CDC25 antibodies shows expression of CDC25A, B, and C in the cytoplasm of luteal cells from the early stage (×200), midstage (×200), and late stage (×200) of the corpus luteum. The early stage of a corpus luteum tissue section stained with preimmune serum shows no signals (×200). 3 Figure 2 Expression pattern of CDC25A protein with advancing stages of CL. Western blot shows an abundant amount of CDC25A protein in early and midstages, compared with the late stage of the luteal phase of the human corpus luteum. Asterisk indicates statistical significance (P<.05). Fertility and Sterility  , DOI: ( /j.fertnstert ) Copyright © 2009 American Society for Reproductive Medicine Terms and Conditions

4 Figure 3 In vivo interaction of CDC25A with DAZL. Protein extract from midstage of CL and GLCs were incubated with DAZL antibody, and immunoprecipitated pull-down proteins were analyzed by Western blot using a CDC25A antibody. Likewise, protein extracts from human CL was incubated with a CDC25A antibody, and immunoprecipitated pull-down proteins were analyzed by Western blot using a DAZL antibody. Control experiments using preimmune serum instead of specific antiserum to DAZL and CDC25A during the co-IP experiments as negative control and cell lysate from CL and GLCs as positive control. IP: immunoprecipitation, IB: immunoblot. Fertility and Sterility  , DOI: ( /j.fertnstert ) Copyright © 2009 American Society for Reproductive Medicine Terms and Conditions
