1 Nociceptive phenotype alterations of dorsal root ganglia neurons innervating the subchondral bone in osteoarthritic rat knee joints  K. Aso, M. Izumi, N. Sugimura, Y. Okanoue, T. Ushida, M. 2 Fig. 1 Representative photomicrographs of CGRP- and TrkA-IR neurons, and FB- and DiI-double-labeled neurons. FB-labeled neurons innervating the subchondral bone of the distal femur are shown in blue (arrows) (A, D) and DiI-labeled neurons innervating the knee joint are shown in red (arrowheads) (B, E). CGRP- or TrkA-IR is shown in C and F respectively. Asterisks indicate CGRP-IR, TrkA-IR and FB (yellow), DiI (red), FB and Dil-double (white) labeled neurons. Left, middle and right are the same section. Figures D and H show negative control for anti-CGRP and anti-TrkA antibody, respectively. CGRP, calcitonin gene-related peptide; DiI, 1,1′-dioctadecyl-3,3,3′,3'-tetramethylindocarbocyanine perchlo-rate; FB, Fast blue; IR, immunoreactive; TrkA, tyrosine receptor kinase A. 3 Fig. 2 Percentage of CGRP-IR DRG neurons innervating the subchondral bone (A) and the knee joint (B) per soma size range. L3 and L4 DRGs of six rats were evaluated. Bars represent 95% CIs. 4 Fig. 3 Representative photographs of histological alterations after MIA injection in the knee joint. The cartilage and subchondral bone in control rats was normal (A, D). At 2 weeks, severe damage of cartilage surface with loss of proteoglycans and chondrocytes (arrows) presented. Mesenchymal change in subchondral bone marrow (double arrow) and increased basophilia at the tidemark (arrowheads) were observed (B, E). At 6 weeks, there were multifocal and fragmentation of subchondral bone with replacement by fibrous tissue (arrowheads). Increased thickening of subchondral bone subjacent to the area of severe cartilage lesion was also observed (double arrowheads) (C, F). Sections were stained with Safranin O and fast green. 5 Fig. 4 The effect of OA induction on behavior tests. (a) Percentage of weight distribution of the ipsilateral hind paw using an incapacitance tester, (b) Mechanical threshold to von Frey filament. Values are mean ± 95% CI. *P = 0.005; vs control. #P = 0.003; vs control. $P = 0.002; vs control. **P = 0.004; vs control. ##P = 0.006; vs control.
