FIGURE 2 Elution pattern of sperm membrane proteins from heparin affinity column, hybridization of heparin-binding sperm membrane protein (HBSM) with [125I]-heparin, and dot-ELISA. (A) Fractions eluted with gradient NaCl concentration stepwise; peak I, II, and III were eluted with 0.25, 0.5, and 1.0 M NaCl solution, respectively. (B) Peak I and II were concentrated and analyzed by 10% (wt/vol) SDS-PAGE with 20 μg of protein per lane. Lane 2, peak I; lane 3, peak II; and lane 1, molecular weight marker. (C) Purified HBSM protein was transferred to nitrocellulose membrane and hybridized with radiolabeled heparin. (D) Dot-ELISA of HBSM. Five micrograms each of HBSM (dot 2 and 4) and 10 mM α-methyl mannopyranoside eluted fraction from ConA column (dot 1 and 3) were dabbed on nitrocellulose membrane and stained with HBSM antisera (dot 1 and 2) and pre-immunized rabbit sera (dots 3 and 4).
