2 Figure 1 Spleen and liver iron load in the C3H/DiSnA, the B10, and the 27 HcB/Dem strains. Results are the means of assays in 9 to 12 male mice. Sixteen RCS (open circles) do not differ significantly from the C3H/DiSnA background strain. Five RCS have similar spleen iron content but either higher (open triangles) or lower (solid triangles) liver iron; 5 RCS have comparable liver iron but lower spleen iron (solid diamonds); and 1 RCS has a higher iron content in both liver and spleen (solid square). Tissue iron loads were analyzed by analysis of variance, and the differences between the strains were evaluated using Dunnett T3 multiple comparison test. 4 Figure 3 Functional consequences of the R435X mutation. (A) Liver Cp mRNA quantifications for 6 inbred strains of mice. We used the comparative Ct method; GAPDH is the reference amplification gene, and the B10 strain is the reference condition. Results are expressed as a percentage of the mRNA value for the B10 strain. Data were obtained from 5 mice in each group. (B) Serum ferroxidase activity was measured in 5 to 10 mice in each group and expressed as a percentage of the activity found in the serum of C3H/HeNrj mice. Mice are identified according to their genotype at the PromCp locus, and Cp+/+ refers to C3H/HeNrj wild-type mice, Cp+/m to C3H mice heterozygous for the R435X mutation, and Cp−/− to C3H/DiSnA CpR435X/R435X mice.
