3 Fig 2 Detection of Merkel cell polyomavirus (MCV) DNA using polymerase chain reaction (PCR) in a Brazilian patient with Merkel cell carcinoma (MCC). The PCR primers were minor modifications of the primer set LT3 (formerly described by Feng et al).3 PCR products were analyzed on 2.0% agarose gel electrophoresis and visualized on an ultraviolet transilluminator. A, Lane M shows the fX174RF DNA marker (Promega Co); lane 1 shows MCC; lane 2 shows MCV-negative control DNA extracted from peripheral blood mononuclear cells (Promega Co); and lane 3 shows the reagent control. The expected 309bp PCR product of MCV can be seen in lane 1. B, Beta-globin reference gene PCR assays. Lane M shows the fX174RF DNA marker; lane 1 shows MCC; lane 2 shows the MCV-negative control DNA extracted from peripheral blood mononuclear cells (Promega Co); and lane 3 shows the reagent control.
