1 Dermal Clusters of Mature Dendritic Cells and T Cells Are Associated with the CCL20/CCR6 Chemokine System in Chronic Psoriasis  Tae-Gyun Kim, Hyunjoong Jee, Judilyn Fuentes-Duculan, Wen Hao Wu, Dashlkhumbe Byamba, Dae-Suk Kim, Do-Young Kim, Dae- Hyun Lew, Woo-Ik Yang, James G. 2 Figure 1 DC-LAMP+ dermal dendritic cells (DCs) and T cells in the psoriatic dermis expressed CC chemokine receptor 6 (CCR6) in the characteristic aggregating pattern. (a) Fraction of HLA-DR+ (left), CD11c+ (middle) dermal myeloid DCs, and a large group of CD3+ (right) infiltrating T cells expressed CCR6 and showed an aggregating pattern. (b) Most dendritic cell-lysosomal-associated membrane protein (DC-LAMP)+ mature dermal DCs in psoriasis co-expressed CCR6 and intermingled with other CCR6+ cells. (c) Flow cytometric analysis of the dermal single cells from the normal and the psoriatic skin revealed that CCR6 expression was confined to the DC-LAMP+ mature DCs (HLA-DR+CD11c+DC-LAMP+ cells), and that immature DCs (HLA-DR+CD11c+DC-LAMP– cells) were devoid of CCR6 (red line: isotype control, blue line: CCR6 stained). White arrows indicate the double-positive cells. Bar=100μm. Journal of Investigative Dermatology  , DOI: ( /jid ) Copyright © 2014 The Society for Investigative Dermatology, Inc Terms and Conditions

3 Figure 2 Myeloid DCs and T cells were the cellular sources for CC chemokine 20 (CCL20) in psoriatic dermis. (a) Dermal CD11c+ DCs did not co-express CCL20 in the normal skin (left); however, psoriatic skin contained some CCL20+ dermal myeloid DCs (right). Note the lack of CCL20 in the normal epidermis, in contrast to the prominent CCL20 staining seen in the psoriatic keratinocytes. (b) CD3+ dermal T cells were devoid of CCL20 in the normal skin (left), but the infiltrating T cells of the psoriatic skin expressed abundant CCL20 (right). White line denotes dermoepidermal junction. Bar=100μm. Journal of Investigative Dermatology  , DOI: ( /jid ) Copyright © 2014 The Society for Investigative Dermatology, Inc Terms and Conditions
