Sildenafil was applied once daily at a dose of 100 mg/kg bodyweight. Placebo-treated mice received 2% methylcellulose. After three weeks of hypoxia maximal walking distance was assessed. Mice that received Sildenafil and performed regular exercise training showed a significantly improved maximal walking distance compared to placebo-treated mice. Data represent mean ± S.E.M. In order to delineate the pathway underlying the effects of regular exercise training on hypoxia-induced pulmonary hypertension we examined the expression of several proteins that are potentially involved in the process, on m-RNA and protein level. Phosphodiesterase (PDE)-5 has been shown to be involved in PAH pathogenesis as demonstrated in numerous publications, and was regulated in response to hypoxia and exercise in our mouse model. Thus, we investigated PDE-5-expression in the lungs of patients with pulmonary hypertension using immunohistochemistry. Phosphodiesterase-5 was significantly upregulated in the lungs of patients with pulmonary hypertension. Figure 1. The treadmill for mice. The treadmill for mice was developed by Arnfinn Sira, Trondheim, Norway. For an initial assessment of their physical performance, the maximal exercise capacity of the mice was tested. The treadmill was sped up to 15 m/s Every three minutes the treadmill was accelerated by 0,05 m/s.
