Immunoblots of (A) Mm5MT cytosolic extracts and (B) extracellular MMTV purified from milk curd in weanling pups stomachs. Serologic reactivity to PBC patient serum and affinity purified AMA from PBC serum was observed to proteins at a molecular weight consistent with the mitochondrial autoantigens PDC-E2 at 74 kilodalton and PDC-E3 binding protein at 57 kilodalton (arrowheads). While incubation with 50 μmol bovine PDC-E2 abrogated reactivity to the 74 kilodalton and 57 kilodalton bands (PDC-E2 +), it had little effect on PBC patient reactivity to gp52 and other characterized MMTV proteins (PDC-E2 -).
