2 Figure 1 TOL proteins are the functional equivalent of ESCRT-0 in plants. TOLs bind ubiquitinated transmembrane cargo proteins, likely through their VHS/GAT domains. The VHS domain might also play a role for recognition of phosphatidylinositide-enriched domains of the plasma membrane. TOLs presumably convey cargo to the ESCRT subcomplexes that will pass the cargo on its way to the vacuole for degradation, but the exact nature of this interaction is currently unresolved.
