Stem Cells BMI: Regenerative Medicine

2 What are stem cells? Undifferentiated cells that give rise to other types of cells. The hope is that these cells can be used to repair tissues and grow new organs.

3 Totipotent stem cells When a zygote first begins to grow these are called Totipotent Stem cells. These can give rise to all the tissues needed for the body as well as cell types needed for the embryonic tissues such as the placenta. As the zygote divides and goes through the various stages of development ,these cells begin to differentiate, or “Told what to do” Once a cell differentiates into another type of cell, it can’t differentiate back into another type of cell.

4 Embryonic Stem Cells These are Pluripotent Stem Cells, which can give rise to all the other types of body cells. Human embryonic stem cells are derived from human embryos created during in vitro fertilization. The embryos are donated for research purposes. These cells are taken during the blastocyst stage. Because these have the potential to save lives but also destroys the embryos, there is a lot of controversy on this subject.

5 Multipotent Stem Cells
Adult stem cells also called Somatic Stem Cells, are undifferentiated cells found in differentiated tissues of children and adults and these are MULTIPOTENT stem cells. They can give rise to the multiple cell types needed in the tissue they come from.

6 Current use Only stem cells found in the bone marrow (hematopoietic stem cells) currently are used in medical treatment. Hematopoietic or Blood stem cells are used to treat blood disorders such as leukemia an lymphoma Stem cells from the patient or donor’s bone marrow replace diseased bone marrow cells

7 Stem Cells Treatments using other types of stem cells are mostly in preclinical stages or early clinical trials. Stem cells may help replace diseased tissue either by integrating with the tissue and producing new cells or by producing growth factors that could cause the patient’s cells to regenerate and repair themselves.

8 Burn patients Researchers are experimenting in re growing skin on burn patients First: skin stem cells are isolated rom an unburned area on the patient’s won skin Skin stem cells and other skin cells are placed directly on the burn A special bandage that provides nutrient fluids and clears wastes are used to aid in healing The stem cells provide growth factors that promote healing

9 diseases In 2012, clinical trials tested the safety of using retinal cells derived from human embryonic stem cells to treat two progressive eye diseases that usually result in blindness. Other clinical trials are testing the use of stem cells to treat heart disease, diabetes and other diseases.

10 Induced pluripotent stem cells
In 2006, Japanese researchers published the first report of induced pluripotent stem cells (iPSCs) in mice. In 2007, that group and two others published results that they had created pluripotent stem cells from adult cells in humans. They did this by using viruses to insert genes for transcription factors into the DNA of various types of cells.

11 Induced Pluripotent Stem cells
This seem to reprogram the cells back to an undifferentiated state. Unfortunately, the reprogrammed cells are not exactly like embryonic stem cells They do not always behave in the same way, they have different epigenetic markers and they sometimes lead to tumors in experimental animals.

12 New discovery Researchers are taking stem cells from amniotic fluid
Amniotic fluid is the fluid that surrounds the developing baby in the womb They are taking the stem cells during amniocentesis or during birth These stem cells are multipotent and can form all sorts of tissues These have not formed tumors in animals
