2 Figure 1 Model for Chromatin Loop Formation Mediated by RNA Polymerases Moving Cohesin Rings along DNA to CTCF-Binding Sites Two RNA polymerases moving in opposite directions drive extrusion of chromatin loops through cohesin rings. The loop extrusion stops when cohesin encounters a DNA site occupied by CTCF on each side of a growing loop. Loop extrusion could be mediated either by a single cohesin ring or by two cohesin rings that embrace the chromatin fiber and are linked to each other either topologically or by complex formation (handcuff model).
