Turn on power of all equipment (microscope, computer, camera Insert 40mL of distilled water into a 6mm-diameter well Load the capture card software Put under microscope Put a worm in the well using a pick Image Processing Separated the video into frames using Windows Movie Maker Using Matlab Import into Matlab Subsampling images Color separation for image segmentation Analysis of the difference between frames Determine the velocity of the worm based on its centroid Record video of the worm using the camera software AVI-analog video file format to image processing

Load AVI-analog video into MatLab Downsample each frame by 50% Color Separation Filter background noise by taking difference between frames Detrend each data plot Plot the components of the centroid x-component Determine worm centroid in each frame y-component Image Processing Separated the video into frames using Windows Movie Maker Using Matlab Import into Matlab Subsampling images Color separation for image segmentation Analysis of the difference between frames Determine the velocity of the worm based on its centroid Apply FFT Numerical and graphical results Determine the frequency at the peak Add both components Determine sinusoidal cycle

6 Detrend Detrending is the statistical or mathematical operation of removing trend from the series It is often applied to remove a feature thought to distort or obscure the relationships of interest In our case, the feature we are removing is the translation of the worm
