Sci ;47(9): doi: /iovs Figure Legend: Elevated pressure induced MG132-sensitive nuclear translocation of NFκB that was sustained in microglia, but not in astrocytes. After 24 hours at ambient pressure, NFκB (green) was weakly expressed in astrocytes (A), but more intensely expressed in microglia (B). For both cells, NFκB was restricted to the cytosol and absent from the nucleus (blue). (C, D) Exposure to elevated pressure for 24 hours substantially reduced cytosolic NFκB and did not induce nuclear localization in astrocytes, but increased cytosolic NFκB and nuclear localization in microglia (white arrowheads). (E, F) Treatment with 10 μM MG132 before and during exposure to elevated pressure for 24 hours does not alter NFκB in astrocytes, compared with pressure alone (C), but may decrease cytosolic NFκB in microglia, as well as preventing nuclear localization (black arrowheads). MG123 did not alter NFκB localization, compared with ambient pressure (insets).
