5 MOUNTANTS DEFFINITION:-
Sections are mounted under cover slip to maintain high refractive index necessary for critical microscopy and to protect the section during storage.

6 TYPES OF MOUNTING MEDIA
Aqueous mounting media Resinous mounting media

7 AQUEOUS MOUNTING MEDIA
It is used for material which is unstained stained fat,metachromatically stained and certain enzymes histochemical methods

8 RESINOUS MOUNTING MEDIA
For routine staining technique where the sections are mounted from xylene.

9 AQUEOUS MOUNTING MEDIA
Most of these media have a low refractive index (1.4 – 1.42),although higher level are obtained with certain use of syrups occasionally single reagents such as pure or dilute glycerin are used. But generally these media are of three types. SYRUPS GELATINE MEDIA GUM ARABIC MEDIA

10 All aqueous media mounting media should contain a bactriostatic agent such as a crystals of thymol,0.25% phenol or 0.025% Na merthiolate to prevent the growth of fungus or moulds.

1.GLYCERINE GELLY 2.APATHYS MEDIUM 3.HIGHMANS MODIFICATION OF APATYS MEDIUM 4.FARRANT’S MEDIUM 5.FRUCTOSE OR LAVULOSE SYRUP’S

12 1.GEYCERINE JELLY Refractive index :1.47
These is usually regarded as a standard mountant for fat stain. Contents :- Gelatin : 10gm dist.H20 : 60ml Glycerin : 70ml Phenol : 0.25gm

13 Dissolve the gelatin in the dist
Dissolve the gelatin in the dist.H2o in conical flask in water bath, using just sufficient heat to melt the gelatin Add glycerin and phenol mix well &transfer the containers. For use melt in a water bath or wax oven. Avoid the a shaking to speed this process, otherwise the mountants will be full of air bubbles

14 2.APATHY’S MEDIUM Refractive index : 1.52
This medium is used when an aqueous medium of higher refractive index is required. It is the use full aqueous mountant for fluorescent microscopy.

15 Contents : Gum Arabic : 50gm Cane sugar : 50gm Thymole : 0.05gm D/w : 50ml

16 3.HIGHMAN’S MODIFICATION OF APATHY’S MEDIUM
Refractive index :1.52 This medium is recommended for use with meta chromatic stage. Contents : Potassium acetate: 20gm Gumarabic : 20gm Cane sugar : 20gm Phenol :0.25gm D/W : 40ml

17 4.FARRANTS MEDIUM Refractive index : 1.43
These medium being liquid is more convenient than glycerin jelly for mounting. but has a disadvantage that is it take much longer to set and air bubbles from during settling process

18 Contents : Gumarabic : 50gm Distilled H2o: 50ml Glycerin : 50 ml Phenol : 0.25gm

19 5.FRUCTOSE/LAEVULOSE SYRUP’S
Refractive index : 1.47 This syrup used as a temporary or special mountant. But it is not recommended for routine use Contents: Fructose : 75gm phenol : 0.25gm Distil.H2o : 25ml

21 RESINOUS MOUNTNG MEDIA
This media are composed of resin (natural and synthetic) either it is natural solvent or dissolved in solvent. Such as xylene until sufficiently liquid to allow easy mounting, compatible with fairly rapid drying and hardening.

22 TYPES OF RESINOUS MOUNTING MEDIA
1.Canada balsom: Refractive index : 1.52 Canada balsom from Canadian fir tree( abies balsamea),is a solid resin and is composed of terpenes,carboxylic acid and their esters; it is usually dissolved in xylene. H&E stain slides are fairly well preseved,but basic aniline dyes tend to fade and Prussian blue is slowly bleached.

23 2.DAMMAR BALSAM It is similar to Canada balsam. It is rarely used today because of the dirt and impurities usually present, and the difficulty of filtering prepared mountant.

24 3.COLOPHANIUM RESIN This is occasionally used as a mountant, when it dissolved in xylene. In alcoholic solution may be used as a differentiating agent

25 3.TERPENE RESIN: In our hands this has proved to be suitable replacement for Canada balsam, being less acid and considerably cheaper

26 SYNTHETIC RESINS There are a great number of synthetic resin either made in the laboratory or prepared commercially. The most commonly used are the polystyrene such as Kirkpatrick and Landrum’s DPX. Plasticizer, such as tricresyl phosphate or dibutylphthalate should be incorporated with the solution of polystyrene in xylene, other wise air spaces appear under the cover slip as the preparation dries.

27 1.KIRK PATRICK AND LENDRUM’S DPX
Refractive index : 1.52 Contents : Distrene : 10 g Dibutylphthalate : 5 ml Xylene : 35 ml

28 This is the most commonly used routine mountant preserving.
It has a great advantage over balsam that slides can be cleaned of excess mountant simply by stripping it off after cutting around the edge of the coverslip.
