3 Excel Calculator – User Instructions
Each gender has a specific calculator Participant’s height will be entered by the user IBW and # of grams of protein required to reach goal of 2.5 g/kg/day will be calculated based on height. Users will next enter protein values from standard nutritional care. Finally, the total amount of study amino acids and the subsequent volume of Clinosol 15% to enter will be calculated automatically based on IBW and protein from Standard nutritional care.

4 Excel Calculator – User Instructions
The maximal rate of infusion of AA is 1.8 g/kg IBW/day The calculator will also calculate the maximum allowable amino acid dose and maximum allowable volume of Clinisol 15% that can be administered. The calculator is programmed to indicate if the value Amino Acid dose and Volume of Clinisol 15 % exceeds the Maximum allowable dose. If the Volume of Clinisol 15% required to meet daily protein goals is greater than the Maximum allowable dose, it will appear in red text and the maximum allowable dose will appear in green text. (See example) Users should administer the amount that appears in green text (See examples)

5 Considerations for Using AA Calculator
The previous 24h protein amount should be calculated and used to generate the new daily amino acid dose. Consider whether the amount of protein via standard of care methods will be the same, less or more than in the past 24 hours when setting the final target for the day. For example, if the patients is planned to receive more enteral nutrition (and thus protein via the enteral route), you may need to adjust downwards the desired amount of IV aa. When you have to make a judgment, consider protein amounts in past days and if the patient has received less than desired, err on side of delivering too much.

Just starting on EN, expected to receive 80 grams protein/day Use ‘Male Participants’ Calculator Enter the height of the man into the ‘Height (cm)’ circled in red below This will automatically calculate the IBW and the Daily Protein Goal

7 Example: Mr. X Participant is receiving 80g of EN protein, so enter this value under the ‘Standard Nutritional Care’ section (circled in red) under the correct source of the protein type (EN) (circled in green) 4. Add the amount of protein in grams under the appropriate source into the calculator so that the correct amount of amino acids (Clinisol 15%) can be calculated to be administered

8 Example: Mr. X 5. The amount of ‘Study Amino Acids’, ‘Amino Acid Dose’ ‘Volume of Clinisol 15% Required to Meet Daily Protein Goals’ and ‘Maximum Allowable Volume of Clinisol 15%’ will automatically be calculated based on input of height (cm) and the amount of protein in grams from ‘Standard Nutritional Care’. Note: For the ‘Volume of Clinisol 15% to be Administered’ (outlined in red), the indicator of whether the volume required exceeds the maximum allowable dose is based on the text colour . If the volume required value is green, use the this value. If the volume required value is red, do not use the value, use the Maximum allowable value, which is now green.

9 Example: Mr. X Summary The volume of Clinisol 15% that will be administered to Mr. X, who is a 55 year old man, 6 feet tall (183 cm) and weighing 204 lbs (92 kg) and receiving 80 g of EN protein is 914 mL. The volume of 914 mL is used (as indicated by its green coloured text) because it does not exceed the Maximum allowable Volume of Clinosol, which is 933 mL in this scenario.

10 Example 2: Mrs. Y 68 year old woman, 5 feet tall (152 cm), 158 lbs (72 kg) Just starting on EN, expected to receive 80 grams protein/day Use ‘Female Participants’ Calculator Enter the height of the woman into the ‘Height (cm)’ circled in red below This will automatically calculate the IBW and the Daily Protein Goal

11 Example: Mrs. Y Participant is receiving 40g of EN protein, so enter this value under the ‘Standard Nutritional Care’ section (circled in red) under the correct source of the protein type (EN) (circled in green) 4. Add the amount of protein in grams under the appropriate source into the calculator so that the correct amount of amino acids (Clinisol 15%) can be calculated to be administered

13 Example: Mrs. Y Summary The volume of Clinisol 15% that will be administered to Mrs. Y, who is a 68 year old woman, 5 feet tall (152 cm) and weighing 158 lbs (72 kg) and receiving 40 g of EN protein is 583 mL. The volume of 583 mL is not used (as indicated by its red coloured text) because it exceeds the maximum allowable Volume of Clinosol, which is 542 mL in this scenario. Instead, 542 ml is used (as indicated by the green coloured text) because it is the maximum allowable dose for this participant.
