2. PGT FOR SICKLE CELL ANEMIA Introduction and Aims
Sickle cell anemia is an autosomal recessive disease resulting from the presence of mutated forms of hemoglobin in red blood cells
In Nigeria, sickle cell anemia affects about 1-3% of the population.
PGT combines ART, molecular and cytogenetics for testing genetic disorders
This study compared the outcome of all cases of PGD for sickle cell anemia using cleavage and blastocyst stage biopsy in a private hospital setup in Nigeria.

4. PGT FOR SICKLE CELL ANAEMIA Methodology
All PGT cycles for sickle cell anemia from April February 2017
Cleavage biopsy- assisted hatching on day 3
Trophectoderm biopsy- assisted hatching on day 3
or morning of day 5
Biopsy done by gentle traction and laser pulsation (ZILOS-tk Laser)
1 or 2 cells removed for cleavage biopsy, 3 to 5 cells for trophectoderm biopsy
Embryos vitrified for subsequent thaw and transfer

5. RESULTS After PGT, the results indicate that:
Cleavage biopsy produces a higher number of ‘No Result’ than Trophectoderm biopsy
Trophectoderm biopsy records a higher number of normal embryos compared to cleavage biopsy
The number of abnormal embryos are relatively the same in both groups
The pregnancy and implantation rates show higher values with trophectoderm biopsy when compared to cleavage biopsy

6. CONCLUSIONS Key findings of this study are:
PGT using trophectoderm biopsy for interrogating embryos at risk for sickle cell anemia proved superior to blastomere aspiration at the cleavage stage
Higher pregnancy rate with trophectoderm biopsy than blastomere biopsy
