1 Animal cell culture introduction and history of acc

2 CELL CULTURE….. ANIMAL TISSUE (CULTURING) PRIMARY CULTURE
In vitro culturing of animal cells, tissue fragment, organ or part of organ under aseptic condition using appropriate media…. ANIMAL TISSUE (CULTURING) PRIMARY CULTURE (SUBCULTURING) either gives FINITE CELL LINE or CONT. CELL LINE

3 MAMMALIAN CELL CULTURE…….
PRIMARY CULTURE : first established culture SUBCULTURING : transfer of cells from spent out media to fresh media Subculturing leads to generation of cells lines Lineage of cells originate from the primary culture is cell strain Types of culture primary culture : cells directly taken from the tissues (explant culture) secondary culture : derived from primary culture.(monolayer culture , suspension culture)

4 THE PROCESS TO CULTURE CELLS….
HARVEST CELLS ISOLATE CELL PLATE THE CELLS ON APPROPRIATE GROWTH MEDIA IN A CULTURE DISH CULTURE CELLS BY PLACING CULTURE DISH IN CELL INCUBATOR

5 VERIFY CULTURED CELL or SUCULTURE CELLS ARE TYPE OF INTEREST TO OBTAIN PURE CULTURE
CELLS ARE THEN READY FOR MANIPULATION AND MODIFICATION FOR EXPERIMENTAL PROCEDURE

6 CULTURE SYSTEMS………. MONOLAYER Anchorage dependent
cells grow attached to substrate cell growth controlled by contact inhibition. SUSPENSION Anchorage independent grow floating free in media growth controlled by density limitation

8 HISTORY…… 1903: Jolly, showed for the first time that cells can survive and divide in vitro. 1907: Ross Harrison , was able to show the development of nerve fibers from frog embryo tissue, cultured in a blood clot. 1913:Carrel introduced strict aseptic techniques so that cells can be cultured for long periods. 1916: Rous and Jones introduced proteolytic enzyme trypsin for the subculture of adherent cells. 1940s:the use of antibiotics & penicillin and streptomycin in culture medium decreased problem of contamination in cell culture.

9 1952 :Gey produced first human cell line , HELA , from cervical carcinoma
1965: Harris and Watkins were able to fuse human and mouse cells by use of virus. 1975:Kohler and Milstein produced first hybridoma capable of secreting a monoclonal antibody. 1982:human insulin became the first recombinant protein to be licensed as therapeutic agent. 1985:human growth hormone produced from recombinant bacteria was accepted for therapeutic use.

10 1980s: Chinease hamster ovary (CHO) cell lines were developed..
In 1996: Wilmut and co-workers successfully produced transgenic sheep dolly through nuclear transfer technique.
