2 Figure Effect of meloxicam on frequency, amplitude, and duration of RU486-induced uterine contractions. Uterine activity was continuously monitored by uterine EMG electrodes from day 125 of gestation. Uterine contractions were identified on the polygraph chart and manually analyzed at different time points. Controls: C1 to C3 (72, 24, and 12 hours before meloxicam/saline infusion); RU486-induced labor-type contractions: R1 and R2 (4 and 2 hours before meloxicam/saline infusion); treatment with meloxicam or saline T0 to T3 (initiation 6, 12, and 16 hours into infusion), T4 to T6 (24, 40, and 44 hours into infusion, saline-infused animals delivered). Time of RU486 injections is marked with arrows (44 ±4 hours before meloxicam/saline infusion and within 12 hours starting the meloxicam/saline infusion). A. Maternal meloxicam administration resulted in a significant reduction in number of contractions per hour, compared with saline-infused animals that went into the labor (P < .001, n = 4). B. Meloxicam infusion significantly decreased amplitude of uterine contractions (P < .001, n = 4). C. Treatment with meloxicam resulted in a significant increase in duration of uterine contractions (P < .001, n = 4).
