Sci ;52(13): doi: /iovs Figure Legend: Relative expression level of VEGF mRNA (a), mRNA of F4/80 macrophage antigen (b), and OPN mRNA (c) in laser-irradiated choroidal tissue of a WT mouse at days 1 and 3 after laser treatment and immediately after treatment. Fifty-four C57BL/6 mice (WT) were used. One eye was used in each animal. Retinas received laser photocoagulation for the induction of experimental CNV under slit lamp microscope observation. At days 1 (16 eyes) and 3 (20 eyes), and immediately after laser irradiation (time 0, 18 eyes), the mice were killed by CO2 asphyxiation. The choroidal tissue was obtained. Choroidal tissue from two eyes was merged into one tube (one RNA sample). Thus, we obtained 9, 8, and 10 mRNA samples at days 0, 1, and 3, respectively. Argon laser irradiation significantly upregulated mRNA expression of VEGF (a) and F4/80 (b) at day 3, or at days 1 and 3, after laser treatment, respectively. Expression of OPN (c) was also upregulated at day 3. Immunohistochemistry in a day 3 sample showed OPN protein expression in the cells in the laser irradiation lesion (d). *P < 0.01 and **P < 0.05 by Tukey-Kramer test.
