Start workshop on time—do not wait for “stragglers” Welcome participants as they enter room Do NOT do participant introductions unless the workshop group is a very small one (less than 10 people). Introduce self and briefly explain teaching experience and current position The activity that you will do today was developed by the Life Sciences Learning Center at the University of Rochester. We found that teachers wanted access to the activities that we developed even after our grant support had ended. To provide for sustained dissemination, we formed a small spin-off company called Science Take-Out. Explain that Science Take-Out is a small company that manufactures and sells hands-on science kits for middle or high school biology students. Quick Guide Safety Student Instructions DISTRIBUTE Flower Forensics KITS—ONE kit, one student handout, and one bag of cut-outs PER PAIR OF TEACHERS For today, you will be working in teams of 2. Kits typically come with ONE student handout in the bag. One teacher should use the student handout that comes in the kit. The other teacher should use the extra copy at the desk. Teachers may make additional copies if their students are working in teams. Notice that each kit contains safety instructions and also a graphic quick guide. Students can refer to the quick guide to identify the materials they will need. We do not make a materials list. Instead, students are encouraged to get what they need as they work. Explain that I’ve used this activity as a “laboratory practical” or “substitute lesson plan” or “just before vacation filler” Some teachers have enlarged and laminated the quick guides to make “placemats” to help students organize kit materials. Explain that the labeling of droppers and tubes allows for reuse—a significant cost savings. Droppers can be rinsed out, or just squirted out. Tubes can be refilled. Turn to this page

5 The owner of a local flower store (Orchid World) has asked you to investigate a possible crime – the theft and illegal cloning of some of his prized black orchids. He has spent many years creating a new strain of rare black orchid that is resistant to insect predators. He recently noticed that three online flower companies are selling black orchids that seem similar to his. He is suspicious that these companies have stolen and are cloning some of his orchids. Ask someone to read aloud. Do you think that the orchids are illegal clones of the Old World Orchid??? How could you tell for sure.

6 Your task: Analyze the evidence to determine if the orchids sold by the online flower companies are clones of the Orchid World company’s black orchids. Paraphrase—What are you going to do with this lab activity? Your Task: Analyze biological evidence to determine if the orchids sold by the online flower companies (orchid A, orchid B, and orchid C) are clones of (identical to) the Orchid World company’s black orchid.

Having a data table that is separate allows students to avoid a lot of page flipping to record data. If you want to, you may ask the students to design their own data table.

8 Part 1: Analysis of Flower Structure
You will compare the structure of the Orchid World orchid with the orchids sold by the online flower company. Ask participants to complete Part 1: Analysis of Flower Structure

9 Part 2: Analysis of Flower Pigments
You will use paper chromatography to compare the pigments present in the four orchid flowers. Chromatography is a method used to separate the plant pigments into a distinct banding pattern of different colors. Your kit contains droppers of the orchid flower extracts that were created by grinding up the orchid flowers. Follow the paper chromatography procedure below compare the pigments of your four orchid samples. Ask participants to complete Part 2: Analysis of Flower Pigments

10  Part 3: Test for Chemical X
Orchid World’s black orchids produce a special chemical (Chemical X) that protects the flowers from being eaten by insect predators. You will test the orchid flower extracts for the presence of Chemical X by mixing a small amount of the extracts with Chemical X Indicator Powder. A fizzing reaction will occur if Chemical X is present in the orchid extracts. Ask participants to complete Part 3: Test for Chemical X

11 Part 4: Compare the DNA from the Orchid Plants
Cloned plants are produced by asexual reproduction. If the online orchids were cloned from the Orchid World black orchid, they should be genetically identical to the original plant. This means that the coded information in DNA molecules of cloned plants should be identical To the Orchid World orchid. You can use DNA samples from the four plants (OW, A, B, and C) to make “DNA fingerprints” for the plants. Ask participants to complete Part 4: Compare the DNA from the Orchid Plants

12 Part 5: Comparing the Protein Amino Acid Sequences from the Orchids
The genetic information stored in DNA is used to assemble amino acids into protein chains. Cloned plants should produce identical proteins. The final test that you will do is to compare the sequence of amino acids in the proteins produced by the flower color gene in the four orchids. Ask participants to complete Part 5: Comparing the Protein Amino Acid Sequences from the Orchids

13 Part 6: Data Analysis You will now analyze ALL of the data that you have recorded on the data table. You may find it helpful to highlight or circle the characteristics that that orchids A, B and C have in common with the Orchid World orchid. Which of the orchids (Orchid A, B or C) are likely to be clones of the Orchid World orchid? Why or who not? Support your answer by citing specific evidence from your data analysis. Ask participants to complete Part 6: Data Analysis If time permits, ask several teachers to read their answers to the questions. If you do this, put the previous slide with the data table on for reference as teachers read their answers.
