1 Essential idea: Enzymes control the metabolism of the cell. Topic 2.5 - Enzymes http://www.hhmi.org/biointeractive/seeing-single-molecules

2 https://www.youtube.com/watch?v=OttRV5yk P7A Proteins These things do stuff! But why? Check for understanding: Why can’t we just rely on normal molecular collision for the reactions to take place?

9 DATA ANALYSIS Generate a graph of enzymatic activity for the enzymes in this simulation for each of the factors listed above. Overlay the graphs on top of one another on the same set of axes. Be sure to keep control variables constant, such as time, amount of enzymes, etc. so that you have meaningful data. Choose a wide range of variables, with consistent increments as well.

20 The Role of Alcohol in the Body – A Metabolic Pathway The liver plays a key role in the breakdown of alcohol (ethanol) in the body. Which enzyme has the product acetaldehyde? Which enzyme has substrate acetaldehyde? If you were to try and stop this metabolic pathway from functioning, how would you do so? Analysis Question:

25 Use the metabolic pathway to the left to answer the following questions: 1.What is the first substrate in this pathway? 2.What is the first enzyme in this pathway? 3.What is the inhibitor of this pathway? 4.What is the end product of this pathway? Check out this awesome visual database of many of the metabolic reactions that take place in the human body. Prepare your brains….they might explode.

27 Practical 3 Enzyme Activity Lab Students will: Design an experiment to test the effect of a factor that affects enzyme activity. Your lab group is responsible for the design of an experiment involving the following: ENZYME – LACTASESUBSTRATE - LACTOSE Lactase is an enzyme that catabolically interacts with lactose. 28 Research Question : What is the effect of Independent Variable on enzyme activity? Choices for Independent Variable – Concentration of Solute in Solution (3) - Scale Temperature of Solution (3) – Hot Plate pH of Solution (3) – pH Strips and Acid/Base Procedure Guidelines – 1.Set up a stock solution of both enzyme and substrate in 400 mL beakers. Enzyme Solution - Dissolve 1 lactase pill in 200 mL distilled H2O. Substrate Solution - Dissolve 5g lactose powder in 200 mL distilled H2O. 2.Expose your enzyme solution to the independent variable chosen. 3.Mix 50 mL enzyme solution and 50mL substrate solution together in a 250 mL beaker. Allow to mix for 15 seconds. Begin timer at the end of the 15 seconds. 4.Record the amount of glucose at 1 minute intervals using the glucose test strips up to 4 minutes. Set glucose test strips out to allow them to dry after testing. Record after 3 minutes drying. 5.Repeat steps 3 and 4 for the remaining independent variables. 6.Record the data for mg/dL glucose in raw data table that will be used to calculate the RATE OF REACTION.
