A MOLECULAR ANALYSIS OF THE HUMAN POLYOMAVIRUSES DNA SEQUENCES IN PROSTATE CANCER PATIENTS REVEALS A HIGH FREQUENCY OF JCV DETECTION: PRELIMINARY RESULTS Cattarino S, Anzivino E, Bellizzi A, Rodio DM, Pietropaolo V, Sciarra A, Gentile V. Department of Obstretics, Gynecology and Urological Sciences, "Sapienza" University, Rome, Italy Department of Health Sciences and Infectious Diseases, "Sapienza" University, Rome, Italy

Background Fattori di rischio correlabili all’insorgenza del TUMORE della PROSTATA Fattori ormonali Fattori dietetici Fattori occupazionali Razza Familiarità INFEZIONI

Background HUMAN POLYOMAVIRUS TCR (transcriptional control region) JCVBKV WW P Q R S URO1 DNA rearrangements impairing BK virus productive infection in urinary tract tumors. Monini et al 1995 CY A B C D E F Mad-1 JC virus DNA is present in the mucosa of the human colon and in colorectal cancers. Laghi et al 1999

to determine the presence of both polyomaviruses genomes (BKV and JVC) in: prostatic tissue, urine, plasma and PBMC samples to determine the presence of both polyomaviruses genomes (BKV and JVC) in: prostatic tissue, urine, plasma and PBMC samples to assess whether a specific TCR configuration and a particular viral genotype were mainly associated with a normal or cancerous prostate cellular environment to assess whether a specific TCR configuration and a particular viral genotype were mainly associated with a normal or cancerous prostate cellular environment Aim of the study

Material and Methods Prostatic tissue specimens, blood, urine samples and PBMC 15 patients (mean age, 66 yr) undergoing radical retropubic prostatectomy (RRP) for PC at our Department (2011-2012) BKV/JCV Real Time TaqMan PCR (Q-PCR) PCR for BKV/JCV TCR and VP1 gene typing region Sequencing of BKV/JCV TCR and VP1 regions

Conclusions Although the sample size in this study is small, our preliminary data demonstrate the presence of JCV DNA in 60% of tumor prostatic tissue samples, confirming the results obtained in a previous study (Zambrano et al., 2002 “T he prostate is a complex habitat where mixed infections with oncogenic DNA viruses frequently occur and opens the discussion to the potential role of these viruses in the cancer of the prostate” ), in which JCV has been defined a common inhabitant of the prostate, and opening the discussion about its potential role in Prostate cancer
