ANTIGEN ANTIBODY REACTION By Dr. Emad AbdElhameed Morad Lecturer of Medical Microbiology and Immunology

ELISA It is an abbreviation of enzyme linked immunosorbent assay. Types: Indirect Direct For the antigen For the antibody

Indirect ELISA A known antigen is fixed to a solid phase (microplate wells). The serum of the patient is then added to the wells and incubated. Washing of the wells is done. Antihuman gamma globulin labeled with a suitable enzyme is added to the wells and incubated. A suitable substrate is added and incubated. Color development in the wells means positive serum.

Immunofluorescence It is another form of immunoassays in which fluorescein labeled antibodies are used. Fluorescein is a dye which emits green fluorescence under ultraviolet light. Types: Indirect IF Direct IF For the antigen For the antibody

Indirect IF A known antigen is fixed to a solid phase (slide). The serum of the patient is layered on the slide and incubated. Washing of the slide is done. Antihuman gamma globulin labeled with fluorescein dye is added to the slide and incubated. The slide is then examined under the fluorescent microscope. Positive fluorescence under the microscope means positive serum.

Radioimmunoassay It is another form of immunoassays in which radiolabeled antibodies are used. The radioactivity is measured by gamma counter.
