INTRODUCTION Bladder outlet obstruction (BOO) one of the causes for overactive bladder (OAB). OAB arises from the functional, pharmacological, and molecular biological changes following the onset of BOO. Of these, changes in various types of receptors present in the bladder have been of increasing interest, and to date, studies have focused on muscarinic receptors. However, little is known about the role of nicotinic acetylcholine receptors (nAChRs). To investigate the role of alpha3 and alpha7 nAChRs (  3 and  7 nAChRs) in the bladder, using a rat model with detrusor overactivity induced by partial BOO. MATERIALS AND METHODS Forty Sprague-Dawley rats were used: 10 were sham-operated (control group) 30 were observed for 3 weeks after partial BOO. BOO-induced rats - further divided into 3 groups Two groups of 10 rats HM group; n=10: received intravesicular infusions with hexamethonium (antagonists for  3 nAChRs) MLC group; n=10: received intravesicular infusions with methyllycaconitine (antagonists for  7 nAChRs) BOO group; n=10 Received only saline infusion The contraction pressure and nonvoiding bladder contractions compared between the control and the three BOO-induced groups. RESULTS The contraction interval the MLC group > the BOO group (P<0.05). Nonvoiding bladder contraction almost disappeared in the HM and MLC groups. Contraction pressure Increased in the BOO group compared with the control group (P<0.05) Decreased in the HM and MLC groups compared with the BOO group (P<0.05). Immunofluorescence staining the  3 nAChR signals increased in the urothelium the  7 nAChR signals increased in the urothelium and detrusor muscle of the BOO group compared with the control group. Western blot analysis both  3 and  7 nAChR levels increased in the BOO group (P<0.05). Interna tional Neurourology Journal 2015;19:12-18

CONCLUSIONS  3 and  7 nAChRs are associated with detrusor overactivity induced by BOO. Furthermore, nAChR antagonists could help in clinically improving detrusor overactivity. Interna tional Neurourology Journal 2015;19:12-18
