5 SUBCULTURE Removing one isolated colony from its original plate and transferring it to a new plate to create a pure culture from a mixed culture. It is important to identify unknown organism.

6 Dilution technique Example- Pour plate, Streak Plate and Spread plate Used to separate and isolate different organisms from one another by decreasing the concentration of original sample and to obtain a pure culture via subculturing

13 OBSERVING PLATES in locations where bacteria are crowded closely together, confluent growth appears. In areas where the bacteria are more spread out individual colonies should appear. Each colony is the result of a single bacterial cell undergoing multiple divisions during the period of incubation. If your plate streaking was done correctly, the initial streaking sector should contain a fairly confluent lawn of bacteria. The third and fourth sectors should contain individual colonies.

15 COLONY  Bacteria form colonies when grown in a solid medium. Bacteria  The colony is actually (and ideally) a group of bacterial clones since they are all derived from only a single parent cell.
