4 How a spectrophotometer works
In short, spectrophotometers work by directing a beam of light through a sample and then measuring either the amount of light that is reflected by the sample, or the amount of light that is transmitted through the sample.

5 Measuring Transmittance
By measuring transmission scientists can quantify the amount of material present in a solution. The amount of light transmitted through a sample gives a quantitative measure of the turbidity of the sample. The more turbid a sample is the less light is transmitted through the sample.
