Laboratory Hardware Part 1 Bioscience, Equipment Unit 6 Centrifuge, Water Baths, and Incubators 8/29/2015 1

Bellwork What cooking method do you use to make hard boiled eggs? What happens to Italian salad dressing when you leave it sitting in the fridge over night? 8/29/2015Property of CTE Joint Venture 2

Objectives 1. Identify, select, and operate incubators 2. Identify, select, and operate water baths 3. Properly load, balance, and operate a centrifuges 8/29/2015Property of CTE Joint Venture 3

Terminology Centrifuge Water bath Incubator 8/29/2015Property of CTE Joint Venture 4

Water Bath No not like the tub at home for a bubble bath! Rather a device that maintains water at a constant temperature. It is used in the laboratory for incubations 8/29/2015Property of CTE Joint Venture 5

How to Use a Water Bath Set the temperature of the water bath either with a dial control or a digital control. At the beginning of a lab or experiment, you should check the water bath to see if it is set at the right temperature and filled with water. Note: Water baths should be filled with distilled water. Never put your hand in the water to test temperature 8/29/2015Property of CTE Joint Venture 6

Thumbs Up / Thumbs Down Is it important to check the temperature of the water bath periodically while you are running an experiment or incubation? Be prepared to answer why 8/29/2015Property of CTE Joint Venture 7

Incubator A device used for growth and maintenance of microbiological cultures or cell cultures. There are various types, sizes and uses for incubators 8/29/2015Property of CTE Joint Venture 8

Our Incubator The incubator in our classroom is a ______________ It will control temperature ranges between ____ and _____. It can hold approximately ____ 7cm petri dishes 8/29/2015Property of CTE Joint Venture 9

Uses for an Incubator Depending on the growth and type of incubator, it is possible to complete various culture grows or biological reactions while controlling temperature, degree of agitation, gas (CO 2, O 2, etc.) availability. On slates: Why is an incubator a better choice for a cell growth compared to a warm room or heat lamb? Give at least three (3) reasons. 8/29/2015Property of CTE Joint Venture 10

Keeping Water in a Glass Keeping Water in a Glass Centrifuge Water Glass 8/29/2015Property of CTE Joint Venture 11

Centrifuge Centrifuge A piece of equipment rotates tube around a fixed axis the process causes contents to separate based on density; denser contents to the bottom of the tube lighter objects to the top. It takes advantage of the concept of centrifugal force. 8/29/2015Property of CTE Joint Venture 12

Using a Centrifuge Safety -Never place flammable liquid in a centrifuge -Always use a balance to verify weight distribution -Use correct size tube -Stay with centrifuge during start up and abort run if abnormal noise or vibration - Do not open until centrifuge has completely stopped 8/29/2015Property of CTE Joint Venture 13

Using a Centrifuge SOP Loading Samples 1. Obtain correct tubes for centrifuge you will be using and check size in centrifuge before filling tubes 2. Dispense samples into labeled tubes, cap securely, and insure tubes are in good condition 3. If needed fill water blanks into clean tubes for balancing. Use a balance and adjust the volume in your blanks to ensure even load distribution. 8/29/2015Property of CTE Joint Venture 14

Using a Centrifuge SOP Cont. 1 4. Inspect the centrifuge chamber prior to use to be sure it is free of any debris, dirt or leaks. Clean if needed. 5. Load samples and blanks into opposing slots or baskets. Ensure the load is fully Balanced. 6. Close chamber lid and ensure it latches properly 8/29/2015Property of CTE Joint Venture 15

Using a Centrifuge SOP Cont. 2 7. Set time, temp. and speed to the desired settings prior to initiating the run. 8. Remain by centrifuge until set speed is reached. If any unusual noise or vibration occurs, stop centrifuge immediately and tell teacher. 9. Close chamber lid and ensure it latches properly 8/29/2015Property of CTE Joint Venture 16

Using a Centrifuge SOP Cont. 3 Retrieving samples 10. When the run is complete, wait for centrifuge to come to a complete stop before attempting to open lid. 11. Clean the interior of the chamber so the centrifuge is ready for the next user. 8/29/2015Property of CTE Joint Venture 17

Identify and describe the uses of the following objects on your slate A B C 8/29/2015Property of CTE Joint Venture 18

Practice With Centrifuge With you lab group you will practice using the centrifuge SOP at your lab bench to separate pond water in the preloaded tubes. 8/29/2015Property of CTE Joint Venture 19

Closure In 12 words or less summarize the most important aspect of todays lesson. 8/29/2015Property of CTE Joint Venture 20
