MICROSCOPES Unit 1 – Intro to Science

Types of Microscopes

Electron Microscope  Uses a beam of electrons to magnify image greater than 1000x (1000 times)  Kills observable cells so not used to view live specimens.

Compound Light  Light passes through lenses to magnify image up to 1000X  Used to observe living specimen.  Most commonly used microscope in biology  Compound because of multiple objectives.

Dissecting Microscope  Used for low magnification of three- dimensional objects- objects larger or thicker than the compound microscope can accommodate.  Magnifies between 10 and 40 times (10x- 40x)

Eyepiece Body Tube Revolving Nosepiece Arm Objective Lens Stage Stage Clips Coarse Focus Fine Focus Base Diaphragm Light Microscope Parts

Microscope Magnification  To calculate the power of total magnification, multiply the power of the ocular lens by the power of the objective.  On our compound microscopes we have a 4, 10 and 40 objective.  The ocular lens (eye piece) magnifies 10 times.  Total magnification when looking in the microscope on the 4 power objective would be what? ________

Microscope Facts  When focusing on a specimen always start by using the scanning objective.  When using high power use only the fine adjustment knob.  The microscope will flip and invert an image  The higher the objective the less of an object you can see  (pay attention to these things when looking at the “e”  When in doubt, ask your teacher!!

Microscope care  Always carry with 2 hands  Only use lens paper for cleaning  Do not force knobs  Always store covered  Keep objects clear of desk and cords
