Introdution The posterior capsular opacification (PCO) remains a frequent complication of cataract surgery ranging from 3.5% to 55.6%, depending on the design and material of the intraocular lens. Studies on improved IOL biomaterial have focused on treatments that are able to potentialize the inhibition of cell adhesionStudies on improved IOL biomaterial have focused on treatments that are able to potentialize the inhibition of cell adhesion One of this treatment the polymerization of the acrylic hydrophobic surface IOLs with polyethylene glycol. This kind of treatment has shown a significant reduction in protein deposition and cell adhesion in the IOL (Lee et al., 2007)

Purpose the ability of cell adhesion in the acrylic IOLs coated with hydrophilic polyethylene glycol (threated group) and uncoated IOLs (control group) in crystalline epithelial cells human culture. To evaluate the ability of cell adhesion in the acrylic IOLs coated with hydrophilic polyethylene glycol (threated group) and uncoated IOLs (control group) in crystalline epithelial cells human culture.

Methods The research was Approval by the Botucatu Medical School Ethics committee. undergoing cataract extraction by phacoemulsification in the Botucatu Medical School. We performed the epithelial cells culture from the anterior capsule of 144 patients undergoing cataract extraction by phacoemulsification in the Botucatu Medical School. Two patterns of cells growth in culture were observed, a dendritic one and a rounded one.Two patterns of cells growth in culture were observed, a dendritic one and a rounded one.

Patterns of cell growth: Round pattern Dendritic pattern Methods

When reaching the semi-confluence, five rounded pattern epithelial cell cultures and five dendritic pattern cell cultures were trypsinizedWhen reaching the semi-confluence, five rounded pattern epithelial cell cultures and five dendritic pattern cell cultures were trypsinized 3,000 cells of each culture was seed on the Hydrophilic IOLs (Ioflex®, Mediphacos, Belo Horizonte - Brazil) untreated and treated with polyethylene glycol, both the samples in triplicate. The treated IOLs were submitted to treatment with polyethyleneglycol (PEG 300) 392°F evaporated to a glass chamber at a pressure of – 0.0045 Torr, using radio frequency energy 5 to 15W for 30 minutes. Methods

IOLs were left at 98,6° F in a humidified incubator with 5% CO2 for 2 hours for cell adhesion to occur. Then nutrient with 20% fetal bovine serum was added, and the cultures kept in the incubator for 24 hours. The IOLs were washed three times with PBS to remove the loose cells. The IOLs were washed three times with PBS to remove the loose cells. Then the IOLs were them trypsinized to detach adhered cells. The number of cells removed from which IOL was counted with hemocytometer. Methods

Overall, there was less cell adhesion on IOLs treated compared to untreated controls, with statistical significance (p <0.05). There was no statistical difference in samples of round pattern; however, there is a tendency for greater adherence in untreated IOLs (p = 0.421).There was no statistical difference in samples of round pattern; however, there is a tendency for greater adherence in untreated IOLs (p = 0.421). Samples of dendritic cells also showed no statistical difference, but with a significant trend toward greater compliance in untreated IOLs (p = 0.095). Results

By using primary cell cultures, it was possible to add a new variable (cell type) to the experiment, which allowed us to get closer to the real situation. The finding of two distinct types of cells was not expected Analysis of the two cell pattern revealed their distinct behavior. Although no statistical difference was found, the dendritic pattern seems to be more adherent in comparison with the round pattern, and the adhesion inhibition in the treated IOLs group was higher in the dendritic pattern in comparison with this control group. Discussion

These findings lead us to suppose that patients with dendritic pattern cells in culture, were more likely to develop PCO because this kind of cells could be transformed into activated myofibroblasts. strainsWe could not find in the literature experiments that cite and compare these two types of cells in culture, since only cell strains were used for this kind of study. Discussion

The coating of the IOLs with polyethylene glycol was effective in inhibiting cell adhesion on treated IOLs when compared to untreated controls (p <0.05).The coating of the IOLs with polyethylene glycol was effective in inhibiting cell adhesion on treated IOLs when compared to untreated controls (p <0.05). The unexpected finding of two patters of human lens epithelial cells in culture opens new perspective for studies on this subject. The unexpected finding of two patters of human lens epithelial cells in culture opens new perspective for studies on this subject.
