2 CRESH PROJECT 3rd meeting of the CRESH project, 22-24 September 2010, Nantes, France Task 7: Tracking juvenile origin with trace elements Introduction : Task 7 programme says that we shall be: "Tracking juvenile origin with trace elements and stable isotope analysis of hard structures (among beaks, gladius, cuttlebone, statoliths)" - stable isotope data will come from task 5 - hard structures are too light for trave elements composition (given the available analysis systems)

3 CRESH PROJECT 3rd meeting of the CRESH project, 22-24 September 2010, Nantes, France Task 7: Tracking juvenile origin with trace elements Introduction : At the University of Caen, scientists specialized in trace elements analysis are in the team "ERPCB" (Equipe de Recherche en Physico-Chimie et Biotechnologies) 2 meetings were organised with this team (May 5th 2010 and July 12th 2010) The contact person for this study is now Mrs. Christelle Caplat

4 CRESH PROJECT 3rd meeting of the CRESH project, 22-24 September 2010, Nantes, France Task 7: Tracking juvenile origin with trace elements What are our objectives : To compare the composition of 4 different samples (4 samples of juveniles caught inshore + 1 recruits) Just a with genetic markers, better discrimination can be achived with a higher number of variables.

5 CRESH PROJECT 3rd meeting of the CRESH project, 22-24 September 2010, Nantes, France Task 7: Tracking juvenile origin with trace elements What trace elements are worth analysing ? + A literature review suggests that the elements that are lost likely bioaccumulating in Cephalopods are Cd, Hg, Ag, Zn, Cu (in decreasing order) Problems with bioaccumulations: - assumes that contamination varies - detoxication processes may occur (not well known in Cephalopods … (S. Seixas and P. Bustamante contacted about this)

6 CRESH PROJECT 3rd meeting of the CRESH project, 22-24 September 2010, Nantes, France Task 7: Tracking juvenile origin with trace elements What methdology ? + The different methods (Mass-Spectrometry) have been discussed → ICP-OES (ICP "optique") qui est moins sensible que l'ICP-MS (dont on ne dispose pas) → SAA-ET(Spectrométrie d’Absorption Atomique avec atomisation électrothermique à l’aide d’un four graphite) qui est plus sensible que l'ICP-OES et multi-élementaire (alors que la SAA-flamme est mono-élementaire par rapport à l'ICP-OES) The main idea is to start with a method that enables to scan a series of elements (ICP-OES) which may reveal differences in other elements than the initial list.

7 CRESH PROJECT 3rd meeting of the CRESH project, 22-24 September 2010, Nantes, France Task 7: Tracking juvenile origin with trace elements What methodology ? + the quantity of material needed for 1 analysis is (1< x < 2g dry matter) this will require pooling specimens + the validation of measures requires certified controls (same taxa ? or same trophic level?) + Literature review under way about differences in contamination levels (within the Channel)

(september - october) + collection of recruits sample (december -january) + processing of the samples
