1 Growth factor gene expression by intimal cells in healing polytetrafluoroethylene grafts

2 Fig. 1 Northern blot analysis of total RNA from an animal 6 weeks after graft implantation, probed for PDGF A, PDGF B, and TGFβ1. Each lane had 10 μg of total RNA; aorta (A), graft intima (G), and newborn rat pup aortic smooth muscle cells in vitro (P). Note the greater expression of PDGF A in graft intima compared to aorta, and the low level of PDGF B expression in both tissues. Rat pup aortic SMCs in vitro express PDGF B and serve as a positive control. In this blot, TGFβ1 expression is slightly higher in the graft intima than in the aorta. Arrows indicate the positions of the 18S (lower) and 28S (upper) ribosomal RNA. 3 Fig. 2 Immunocytochemistry of SMCs in culture. A, Positive staining with smooth muscle actin-specific monoclonal antibody HHF-35; B, Lack of staining with a monoclonal antibody for the endothelial cell marker factor VIII related antigen. 4 Fig. 3 Light micrograph of SMCs in culture. A, Note that the SMCs derived from aorta are confluent, whereas B, the SMCs derived from graft intima are forming nodules and do not reach confluence. 5 Fig. 4 Northern blot analysis of total RNA from SMCs cultured from aorta (A) and graft intima (G1) and (G2) 6 weeks after graft implantation. The blot was probed for PDGF A, PDGF B, and TGFβ1. Note that all the isolates exhibit the capacity to express the three mRNAs, but that the levels of expression are variable. Arrows indicate the positions of the 18S (lower) and 28S (upper) ribosomal RNA.
