The staining was scored as follows: 0=no staining in any area (white section of the column), 1=localized staining in the epidermis surrounding hair infundibulum in 1–2 follicles (light gray section of the column); 2=localized staining in the epidermis surrounding hair infundibulum in less than 50% of follicles (dark grey section), 3=localized staining around hair infundibulum in more than 50% of hair follicles and/or in the interfollicular epidermis (black section). 5 Figure 4 Hyaluronan concentration and hyaluronan synthase and CD44 mRNA levels in tape stripped mouse epidermis. (a) Mouse tail skin was repeatedly tape stripped, and specimens were taken after 1–10 d. Epidermis was separated from dermis with ethylenediamine tetraaceticacid, and the hyaluronan content was measured using an enzyme-linked sorbent assay. The data represent means and SE of untreated controls (day 0, n=40), and tape-stripped animals (day 1, n=2; day 2, n=6; day 3, n=8; day 7, n=3; and day 10, n=3). Analysis of variance showed that the stripping caused a statistically significant increase in the epidermal hyaluronan content (p<0.001), day 3 value being significantly higher than control or day 10 value (Scheffe's test, p<0.001 and 0.01, respectively). (b) RNA was extracted from the epidermis of E18 fetal, newborn, and adult mouse, and adults 3 d after tape stripping. RT-PCR was performed with primers specific for the different has isoforms, and CD44, and related to the band density of glyceraldehyde phosphate dehydrogenase (GAPDH). has1 did not produce a significant signal in any of the samples. The data represent means and ranges from two to five specimens.
