1 Ovarian stimulation modulates steroid receptor expression and spheroid attachment in peri-implantation endometria: studies on natural and stimulated cycles  Joyce Chai, M.R.C.O.G., Kai-Fai Lee, Ph.D., Ernest H.Y. 2 Figure 1 Quantitative PCR analysis of estrogen receptor (ER) α, ERβ, progesterone receptor (PR), and glucocorticoid receptor (GR) transcripts of human endometrial samples from natural (N) and stimulated cycles, in which those with serum E2 ≤20,000 pmol/L on the day of hCG injection were grouped as moderate responders (M) and those with a serum E2 level >20,000 pmol/L were grouped as excessive responders (E). ∗Significant difference between groups (P<.05). 3 Figure 2 Immunohistochemical staining of ERα, PR, and GR of human endometrial samples from natural and stimulated cycles. The brown staining of ERα, PR, and GR was found mainly in the nuclei. GE = glandular epithelium; SC = stromal cells; other abbreviations as in Figure 1. 4 Figure 3 The effect of hormones on Jeg-3 spheroid–Ishikawa endometrial cell coculture assay. (A) The concentrations of E2 and P used in the coculture study are shown. (B) Jeg-3 spheroids of sizes 60–150 μm were selected and cocultured with (P4 or E2) or without (Neg) hormone-treated Ishikawa cells. Hormonal concentrations that resemble the serum levels of patients in natural cycles (N) or moderate responders of a stimulated cycle (M) do not affect spheroid attachment in vitro. However, hormonal concentrations that resemble those of excessive responders of a stimulated cycle (E) significantly down-regulate spheroid attachment onto Ishikawa cells in vitro. Methotrexate (MTX, 5 μmol/L) significantly down-regulates spheroid attachment. ∗Significant difference from the control (P<.05).
