1 Contribution of different bone marrow-derived cell types in endometrial regeneration using an irradiated murine model  Claudia Gil-Sanchis, Ph.D., Irene Cervelló, Ph.D., Satish Khurana, Ph.D., Amparo Faus, B.Sc., Catherine Verfaillie, M.D., Ph.D., Carlos Simón, M.D., Ph.D.  Fertility and Sterility  Volume 103, Issue 6, Pages e1 (June 2015) DOI: /j.fertnstert Copyright © 2015 American Society for Reproductive Medicine Terms and Conditions

2 Figure 1 Histomorphological characteristics of recipient uterus. The seven types of bone marrow-derived cells (BMDCs) are summarized in two groups: fluorescence-activated cell sorter (FACS)-sorted BMDCs (A–D) and BMDC lines (E–G) (see upper boxes). Excised uteri were macroscopically analyzed and microscopically by a hematoxylin and eosin (H & E) staining determining the endometrial composition for (A) unfractionated bone marrow (UBM), (B) hematopoietic progenitor cells (HPCs), (C) endothelial progenitor cells (EPCs), (D) mesenchymal stem cells (MSCs), (E) mOct4+ BM-HypoSCs, (F) mOct4− BM-MAPCs, (G) MSC line, and (H) control uterus (physiological conditions). Table in lower panel describes quantitatively the presence of glands per area (in pixels) and the measurement of the endometrial thickness (in microns) in the seven types of BMDCs. 3 Figure 2 Presence and localization of green fluorescent protein (GFP) cells from bone marrow (BM)-derived cells in endometrium. (A) Immunohistochemistry (3,30-diaminobenzidin) for GFP demonstrating the presence of GFP+ cells in the endometrium of the female-irradiated recipients. (A1) GFP+ expressing cell in the stromal compartment. (A2) GFP+ expressing cells in the endothelium of blood vessels. (A3) GFP+ cells in the epithelial compartment. Respective controls are included as the uteri from GFP (control+) and non-GFP (control−) mice. The quantification of engrafted GFP+ cells is represented as percentages of mean ± SEM among the different cellular endometrial compartments (stroma, epithelium, and vessels). (B) Diagram summarizing the contribution of the bone marrow-derived cells (BMDCs) subtypes to endometrial regeneration in a TBI murine model to the different endometrial compartments: epithelium (Epit), stroma (Str), and endothelium (End). Ranges of percentages are represented as: (−) 0; (+) 0.1%–0.4%; (++) 0.4%–0.7%; (+++) 0.7%–1%, and (++++) ≥1%. 4 Figure 3 Proliferation index by Ki-67 and apoptotic index by TUNEL assays. (A) Ki-67 antigen expression is represented for all kind of treated uterus. Images are represented at ×40 magnification. Quantification for Ki-67+ cells versus total counted cells is represented as percentages of mean ± SEM. (B) TUNEL assay is represented for all treated uteri. Quantification for TUNEL+ cells versus total counted cells is presented as percentages of mean ± SEM. *Denotes statically significant differences between the seven bone marrow-derived cell populations. 5 Figure 4 Colocalization green fluorescent protein (GFP)/TUNEL/Ki-67 assays. (A) GFP (green) and TUNEL (red) labeling. Presence of double positive cell is indicated by white arrow. (B) GFP (green) & Ki-67 (red) labeling. The double positive cells are indicated by white arrows. (C) GFP mouse uterus as positive control for GFP (green). (D) Cryopreserved-thawed bovine ovarian tissue as a positive control for TUNEL staining (red). (E) Mouse small intestine as positive control for Ki-67 antigen (red). In all cases, merged images are represented at ×40 (GFP or TUNEL) or ×100 magnification (Ki-67), with nuclear staining represented with 6-diamino-2-phenylindole (DAPI) (blue).
