Ras family G-proteins such as Rheb (orange) bind to PDEδ using a farnesylated cysteine residue at the C terminus that is shown in ball and stick representation (left). The lipid molecule is buried deep within the core of the β sandwich of PDEδ. Zimmermann and coworkers have developed benzimidazole-based compounds that disrupt the interaction between PDEδ and K-Ras by binding in the cavity of PDEδ (right).
