Flow Cytometric Analysis of Thymocytes from pTα+ and pTα−/− Mice
(A) Total thymocytes from (left) pTα+ and (right) pTα−/− mice were surface-stained with phycoerythrin-conjugated CD25 antibodies, and cytoplasmic staining was performed with panTCRβ antibodies. Cells from pTα+ mice were enriched for the CD4−CD8− subset by negative depletion of CD4/CD8 positive cells using Dynabeads prior to staining. Small TCRβ+ CD25+ cells were sorted according to the indicated gates using a FACS Vantage cell sorter. Since the staining was performed on different days, the difference in the CD25 staining between pTα+ and pTα−/− cells is exaggerated.
