During ex vivo chondrogenesis in pellet culture, hMSCs previously cultured with high-glucose medium in expansion culture are less responsive to chondrogenic induction. The high-glucose exposure decreases PKC activity, thus downregulating the expression of TGFβRII in cell pellets. The reduced TGFβRII expression results in decreased TGF-β signaling upon the activation of TGF-β ligand, further leading to reduced chondrogenesis.
