Proliferation history of aged HSC in vivo. Young (2-3 months old) and old C57BL/6 mice (20-24 months old) were injected with NHS-biotin in vivo. 7 or 14 days after injection, BM cells from each individual mouse were isolated and analyzed as described (Nygren & Bryder, 2008). For each individual mouse, the estimated number of cell divisions was calculated as shown in Figure 1B. Presented is the indicated number of cell divisions of candidate HSCs (LSKIL7Ra-Slamf1+) 7 (top panels) and 14 (bottom panels) days post-injection with NHS-biotin of each individual mouse analyzed. x-axis shows the distribution of the proliferation history (0 to 5+ cell divisions; colored bars) of HSCs from each indicated individual mice. y-axis represents the percentage of cells with the indicated proliferation history. Note that HSCs in one aged mouse (Old 6) showed evidence of extensive proliferation, highlighting the caveat of pooling cells from mice for proliferation analyses.
