Consequences of Fas-ligand and perforin expression by colon T cells in a mouse model of inflammatory bowel disease  Stephen J. Simpson, Ype P. De Jong, Samir A. 2 Fig. 1 T cells from the colon of BM→Tgϵ26 mice are potent CTLs. Redirected cytotoxic T cell assays were set up using colon T cells against 51Cr-labeled Fc-receptor positive P815 cells. Effector cells were incubated with targets at different effector to target (E:T) ratios in the presence or absence of anti-CD3 antibody and harvested after 6 hours. Killing in the absence of antibody was <5% maximum in all cases (not shown). Effectors cells comprised (A) IELs or (B) LPLs extracted from the colons of BM→Tgϵ26 mice (▴) or wt (C57BL/6 × CBA/J)F1 mice (●). Data are representative of at least five different assays. 3 Fig. 2 Target cell death is mediated by FAS-dependent and FAS-independent mechanisms. Colon T cells were set up in redirected assays against FAS-expressing targets (L1210F+) and FAS− targets (L1210F−) either in the presence or absence of 6 mmol/L EGTA and harvested after 6 hours. Cell death at E:T 40:1 is represented. Each point represents an assay from a single individual, and lines connect effector cells from the same individual. 4 Fig. 3 Measurement of weight loss and colitis. (A) The percentage of initial body weight was calculated by dividing the final body weight of the animal at week 4 by the initial body weight (measured at time of transplant). (B) The extent of gross colitis (colon thickening and diarrhea) was determined by necropsy (weeks 4-5 after transplantation). Represented are the mean scores ± SEM on a scale of 0-5, calculated according to the criteria outlined in Materials and Methods (*P < 0.05 relative to controls). (C) Histological colitis was determined by examination of sections taken from distal large bowel. These were scored according to the parameters of bowel wall thickening and inflammatory cell infiltrate, as described in Materials and Methods. The combined scores are measured on a scale of 0-9. ○, Animals with significant gross colitis (scores of 2-5); ▵, animals with mild or no significant gross colitis (scores of 0-1). 7 Fig. 4 Histological examination of colons from BM→Tgϵ26 mice. H&E histology of distal colon from BM→Tgϵ26 mice. (A) Severe colitis with a high number of inflammatory cells (3) (see Materials and Methods) and extensive crypt elongation (3) from a PFPnull→Tgϵ26 mouse. (B) Healthy colon with many goblet cells and no inflammatory infiltrate (0), PFPnull→Tgϵ26 mouse. (C) Moderate mucosal inflammation (2) without bowel wall thickening (0) in a PFPnull × gld→Tgϵ26 mouse (original magnification 100×). 8 Fig. 5 Colon CTLs in (A) F1→Tgϵ26, (B) PFP→Tgϵ26, and (C) gld→Tgϵ26 mice. The CTL activity of T cells from colons of gld→Tgϵ26 PFPnull→Tgϵ26 mice with significant colitis was measured, as described in Figure 1. Responses against L1210+ (■) and L1210− (♦) targets were determined, as described in Figure 2. Assays are representative of two separate experiments.
