MICROSCOPES

Microscope Parts: How to focus your microscope… Use COARSE focus 1st. Coarse focus - rough focus, larger knob. Use FINE focus 2nd. Fine focus - used to fine-tune the focus. Smaller knob.

Eyepiece lens (ocular lens) - the lens at the top that you look into. Has a standard magnification of 10X.

Objective lens - the lens closest to the object. There are generally 3/4 of these…all 4 have different magnifications. Low, Medium, High, Oil (we don’t use). You can only use ONE objective at a time. You always want to start looking at something on LOW power objective. Therefore, there are TWO total lens SYSTEMS. #1: eyepiece #2: objectives Nosepiece - revolving nosepiece - holds the objective lens. You turn this to turn to the objective you wish to use. Do not turn by pulling on the objective lens! Do not touch the end of the objectives where the glass is!!!

How do you find the total magnification of microscope???? Multiply the power of the eyepiece lens by the power of the corresponding objective lens. Example Eyepiece 10X (always the same) Objective 5X (can change) Total Magnification = 5 10 = 50X When you see “X” “X” after a number…it means “times”.

When you look through a microscope…what you see is called field of vision. We represent this in circle. When you move from low power to medium/high power, you will LESS of the object but better and closer in. Therefore you will see more QUALITY and less QUANTITY.

Pointer -when you look through eyepiece, you may see a pointer (looks like a toothpick). This is used as a point for reference.

Stage-flat plate that holds the slide/specimen and also consists of stage clips that hold the slide in place. Arm - side of microscope used to carry microscope. Base - bottom support, also used to carry microscope.

Diaphragm/Iris (condenser) - generally a five- hold disc located under the stage. By turning it, you can vary amount of light passing through the stage opening. (can be different #/sizes of holes)

Body Tube – relfects image (upside down).

Microscope Lingo…. Resolution - the ability of a lens system to show fine details. Very clear and focused!!!

Types of Microscopes: Compound Microscope - microscope that has two lens system (objective & eyepiece) Monocular Head Microscope - a microscope head with a single eyepiece Binocular Head Microscope - a microscope with TWO eyepieces…one for each eye!!

MONOCULAR

BINOCULAR

Microscope Rules: 1. Always carry the microscope with two hands….one under base and one on the arm. 2. Never scoot the microscope across a table. Pick it up!! Be careful at all times. 3. Always leave the microscope on low-power (shortest objective lens) and make sure you turn the light off!!! Remember these two things! 4. When first focusing….start on low power and use coarse focus first!! 5. Do not touch glass of microscope.
