A requirement for access to the tissue and a high signal-to-noise ratio, is a short single stranded sequence of high Tm-value of the probe : target duplexes. The higher Tm, the more duplexes at high stringency, the higher specificity. Our sophisticated design algorithm has recently been improved, to narrow the Tm-value even further, aiming at 84 o C. This new probe performance means more probes can be used with the same protocol, enabling ISH Automation and multiplexing. This seminar will take you through important LNA TM ISH optimization tips & tricks, followed by a true clinical story of miR-21 ISH used in colon cancer patient stratification for adjuvant chemotherapy. The large clinical study was follow by miR-21 qPCR validation and launch of a miR-21 RUO kit.

TIP: Using different slide layouts in a presentation gives you flexibility and makes your presentation visually interesting and more appealing for your audience Exiqon Biomarker Seminar MicroRNAs constitute a class of short RNAs which function as post-transcriptional regulators of gene expression. The expression patterns of microRNAs can accurately classify discrete tissue types and specific disease states which have positioned microRNAs as promising new biomarkers for diagnostic and prognostic applications, particulary in cancer. Exiqon Service launched our Biofluid NGS platform last year, using Illumina instruments and sequencing kits. During development our NGS scientists have optimized RNA extraction efficiency, library preparation and QC procedures using > 50 spike-ins to control and monitor sample quality and read quality. This highly robust and sensitive workflow allows profiling of urine, CSF and serum- plasma samples at high quality. This presentation will explain the workflow in a Biofluid NGS project and how we monitor pre- analytical variables as RNA extraction efficiency, presence of undesired inhibitory components and hemolysis, which all affects the microRNA profile. Second part of the presentation will describe how Exiqon uses our sensitive LNA™-based qPCR platform for validation of NGS data. In Sept. 2014, a microRNA QC study performed on serum- plasma samples was published in Nature Methods (Mestdagh et al) showing Exiqon LNA TM qPCR as the only platform being concordant with Illumina and IonTorrent data. This correlation is of outermost importance when you set the criteria for selection of microRNA targets to validate; how low can the expression level be ? - and still keep accuracy, robustness and specificity in another cohort. ●
